A hacker turned off my Trend Micro virus protection. I checked and my program was still in my computer, but I can't bring it up to check it out. What should I do to re-enable my Trend Micro program?

Persons unknown installed a virus on my computer while I was using it. It locked with a message to call a phone number shown on the screen, asking me to pay $249.95 by credit card to remove the virus and free up my computer. I have Stop Sign anti-virus protection but it did not catch this threat. What can I do at this point? Can you provide help?
